What the best way to get to Buffalo from Ithaca?

Take NY-96 north to NY-14. Take NY-14 north for a very short time, where you hit I-90 (Thruway). Take that west to Buffalo. The Thruway toll is $2.90 through exit 50, another 50ยข if you get on I-190 north at I-90.

This is what my atlas says. Toll figures come from the Thruway website. Buffalonians, feel free to suggest a better route (or correct any mistakes).

That's the quickest (and easiest) way.
Be VERY CAREFUL of county sherriffs on Rt. 96. I always see one when I go home, and I do not let my speed go much above 55. They will give you a ticket if you speed even the slightest amount.
the only policeman on 96 ive seen is going thru trumansburg and interlaken, so be sure to slow down thru the villages
